The Spanish outfit took a three-goal lead into the second leg, after winning 4-1 at the Camp Nou last week.
And Ernesto Valverde erred on the side of the caution, playing a full-strength side in Rome to ensure his side progressed.
But incredibly, the Italian side pulled off a stunning 3-0 win at the Stadio Olympico to dump the Catalan side out.
Edin Dzeko opened the scoring after just six minutes to send Roma on their way.
Before captain Daniele De Rossi scored from the penalty spot after the interval.
And as Barcelona attempted to hit back, the Italian side secured their passage to the semi-final when Kostas Manolas netted with eight minutes remaining.
Roma advanced on away goals.
